Yakuriguchi Station opened on 1 September 1961 as an added stop on Japanese National Railways' Kōtoku Line. Initially a flag stop bypassed by many trains, it transferred to JR Shikoku at the 1 April 1987 privatisation and received station number T21. On 14 March 1998, as part of Kōtoku Line speed-up works, the station was relocated some 300 m further from Takamatsu and a passing loop installed, giving it two opposed side platforms in place of the original single platform. The unstaffed station sits 12.3 km from Takamatsu; only local services call. Each platform has a shelter and a small ticket-vending kiosk, with no direct interior connection between them.

Yakuriguchi's name refers to Yakuri-ji, the 85th temple of the Shikoku pilgrimage, which lies about 2 km from the station along the historic Sanuki Kaidō.
