Saikū Station opened on 27 March 1930 as part of the Sangu Express Electric Railway's extension between Matsusaka and Gekū-mae (today's Miyamachi). On 15 March 1941 the line merged with Osaka Electric Tramway to form Kansai Express Railway, then merged again with Nankai Railway on 1 June 1944 to create Kintetsu. A new station building was completed in 1992. PiTaPa support began on 1 April 2007, and the station became fully unattended on 21 December 2013. A second "Historical Park" entrance on the platform 1 side, complete with rest area and tourist information for nearby Meiwa, was added on 19 March 2015.

On 26 March 2011 the station served as a filming location for the NTV Saturday drama The High-School Restaurant, where it appeared under the fictional name "Aikō Station."
