Motohasunuma Station opened on 27 December 1968 with the inauguration of Toei Subway Line 6. On 1 July 1978 Line 6 was rechristened the Mita Line. Originally planned under the working name 'Hasunuma', the station was given the present name because the Tokyu Ikegami Line — then expected to share through-running — already had a Hasunuma Station in Ota Ward. Routine business was handed to a contractor on 1 March 2003 and PASMO usage began on 18 March 2007. A new Entrance A4 and an additional ticket gate on the Izumi-Kōen side opened on 1 April 2010, and direct operation by the Bureau of Transportation resumed on 1 June 2013.

Although Motohasunuma sits underground, the next station to the west (Shimura-sakaue) lacks turn-back facilities, so when weather closes the elevated section of the Mita Line beyond it Motohasunuma sometimes becomes the temporary terminus.
