Mitsukoshimae Station opened on 29 April 1932 as the southern terminus of the Tokyo Underground Railway's line from Asakusa, becoming a through stop on 24 December the same year when service extended to Kyōbashi. The 1941 Land Transport Adjustment Law transferred the line to the Teito Rapid Transit Authority. Hanzōmon Line platforms opened on 26 January 1989 as the line's terminus, becoming through platforms with the 28 November 1990 extension to Suitengūmae; they sit 27.8 metres below ground, dug by roof-shield. The station passed to Tokyo Metro at the April 2004 privatisation, PASMO support began on 18 March 2007, and departure melodies arranged from the folk song "Oedo Nihonbashi" entered service in 2018.

Mitsukoshimae is Japan's first railway station to carry a private company's name in its title — Mitsui Bank executive Shigeaki Ikeda persuaded the Mitsukoshi department store to underwrite the station so it could connect directly to the shop floor.
