Ajisu opened on 17 August 1924 as Hon-Ajisu Station (本阿知須駅), the terminus of the Ube Railway's extension northwards from Tokonami. The line was pushed on to Ogōri (today's Shin-Yamaguchi) on 26 March 1925 and the station became a through stop. The Ube Railway was nationalised on 1 May 1943 and the route was renamed the Ube East Line, then the Ube Line on 1 February 1948. The station was renamed Ajisu on 1 June 1950, taking the name simultaneously surrendered by today's Hon-Yura Station on the San'yō Main Line. Freight handling ended in 1962, parcel handling in 1984. The 1 April 1987 JNR privatisation gave control to JR West.

On 14 July 2001 a temporary Midori-no-Madoguchi staffed ticket office was installed inside the station for the duration of the Yamaguchi Kirara Expo, which used the station as the nearest rail access point for the fairground.
